It is a common component of tobacco smoke and exposure is associated with an increased risk of cancer in smokers. The association of exposure to naphthalene and human cancers is not well established. One mechanism of carcinogenesis in response to exposure to naphthalene is the formation of DNA adducts and the subsequent mutation of oncogenes. The objective of this study is to determine the contribution of naphthalene-DNA adducts to naphthalene-induced mutations. To do this, we will compare the mutagenicity of naphthalene-DNA adducts to the mutagenicity of DNA adducts induced by 2-acetylaminofluorene, a common food mutagen. Specifically, we will 1) isolate DNA adducts formed in vitro from naphthalene; 2) examine the reactivity of the isolated naphthalene-DNA adducts with DNA polymerases alpha, beta and gamma; 3) compare the mutagenicity of naphthalene-DNA adducts to that of the 2-acetylaminofluorene-DNA adducts in human adenovirus T-antigen-expressing and human colon cell line HCA-7 and 4) examine the mutagenicity of naphthalene-DNA adducts in vivo in Salmonella typhimurium. Information gained from this study will contribute to our understanding of the mechanisms of naphthalene-induced carcinogenesis.Share this:
